2D or 3D electron microscopy morphometric analysis of the central nervous system with Deep FIB-SEM and IA-based image analysis
We combine latest developments in cryo sample preparation and imaging by electron microscopies (cryo-FIB/SEM and cryo-TEM) to observe biomolecules of interest (ribosomes, carboxysomes, viruses, nuclear pore complexes…) within their cellular or tissular environment. Using cryo-electron tomography followed by image analysis, we are able to determine their 3D structures with near atomic resolution. Altogether, these methodologies pave the way for in situ structural biology studies.
Gene expression, DNA damage, DNA structure and DNA-binding proteins structures, super-resolution (SMLM, RIM, SIM, Airyscan), single particle tracking, photo-manipulation, Correlative Light Electron Microscopy (CLEM)
Atomic force microscopy on living cells or 3D samples is a powerful technique to measure the mechanical properties such as elastic moduli , rigidity, adhesion force, adhesion energy. Combining optical images, AI and AFM, cell population’s mechanome becomes accessible. 3D samples (tissues, organoids) mechanical properties require specific expertise we also provide.
To decipher Inflammatory and cardiovascular diseases, multiplex imaging and cytometry pipelines include the entire process from sample preparation, image acquisition to image analysis and quantification. Our workflows are adpated to mouse or human lymphoid organs and blood . They employ techniques such as confocal spectral imaging and cytometry-based analysis (spectral, sorting, and analysis).
To tackle complex biological questions in aging and geroscience, we design and implement advanced multimodal, multiparametric, and multiscale pipelines in imaging, flow cytometry, and image flow cytometry. Our workflows span the entire process—from sample preparation to data acquisition and multidimensional analysis. We investigate biological systems across multiple scales, from subcellular structures such as mitochondria to organoids, whole organs, and even entire organisms. Our pipelines are tailored to a wide range of tissues, including adipose tissue, muscle, bone marrow, skin, blood, and periodontal tissue. In addition, our platforms provide access to complementary techniques such as second- and third-harmonic generation (SHG, THG), spectral confocal imaging, high-content imaging, image-based cytometry, conventional and spectral flow cytometry, and mechanobiological analysis. These integrative approaches are designed to support comparative and translational research across multiple species, including fish (with an emphasis on the African turquoise killifish as a model of aging), drosophila, mice, and humans.
Inflammatory and cardiovascular diseases are complex, multifactorial conditions that affect multiple organs and cellular components. To tackle these intricate biological challenges, we employ and advance multi-modal and multi-scale imaging and flow cytometry pipelines. These pipelines include the entire process from sample preparation, image acquisition to image analysis and quantification. Our workflows are adpated to various tissues, including the heart, kidneys, blood vessels, muscles, liver and adipose tissue. They operate across scales ranging from whole organs to subcellular levels, employing techniques such as widefield imaging, confocal spectral imaging, high-resolution imaging (Airyscan), high-content imaging (Axioscan 7, Opera Phenix), and cytometry-based analysis (spectral, sorting, and analysis).
