To tackle complex biological questions in aging and geroscience, we design and implement advanced multimodal, multiparametric, and multiscale pipelines in imaging, flow cytometry, and image flow cytometry. Our workflows span the entire process—from sample preparation to data acquisition and multidimensional analysis. We investigate biological systems across multiple scales, from subcellular structures such as mitochondria to organoids, whole organs, and even entire organisms. Our pipelines are tailored to a wide range of tissues, including adipose tissue, muscle, bone marrow, skin, blood, and periodontal tissue. In addition, our platforms provide access to complementary techniques such as second- and third-harmonic generation (SHG, THG), spectral confocal imaging, high-content imaging, image-based cytometry, conventional and spectral flow cytometry, and mechanobiological analysis. These integrative approaches are designed to support comparative and translational research across multiple species, including fish (with an emphasis on the African turquoise killifish as a model of aging), drosophila, mice, and humans.
Organ: Bone marrow
Inflammatory and cardiovascular diseases are complex, multifactorial conditions that affect multiple organs and cellular components. To tackle these intricate biological challenges, we employ and advance multi-modal and multi-scale imaging and flow cytometry pipelines. These pipelines include the entire process from sample preparation, image acquisition to image analysis and quantification. Our workflows are adpated to various tissues, including the heart, kidneys, blood vessels, muscles, liver and adipose tissue. They operate across scales ranging from whole organs to subcellular levels, employing techniques such as widefield imaging, confocal spectral imaging, high-resolution imaging (Airyscan), high-content imaging (Axioscan 7, Opera Phenix), and cytometry-based analysis (spectral, sorting, and analysis).
With the help of the IBDM Lignt Microscopy Facility we can offer an comprehensive CLEM Pipeline from live fluorescence imaging to EM imaging (in 2D and 3D)
As Volume EM techniques we offer Electron Tomography and Serial Blockface Scanning Electron Microscopy. The targeting of a precise ROI can be achieved by integrating Light Microscopy or X-ray imaging in the pipeline.
High speed, low photobleaching with sub cellular resolution and high detection sensitivity for cleared whole organs, brains or small embryos is offered by the Zeiss light Sheet LS7 -Multiphoton confocal system also available depending on specific applications (including Second Harmonic Generation), large field of view spinning disk (Leica microscope equipped with Yokogawa CSU-WI scanning head) also available and can be used for living sample. Both multiphoton confocal and spinning disk are equipped with 20x, 0.95 NA Water IMM objective providing 2.2 mm free working distance allowing in depth imaging with an excellent 3D resolution in tissue, small animal models, organs, organoids…
Following acquisition, solution or assistance for 3D image analysis, segmentation and quantification can be provided using dedicated software (Imaris, Cellpose, Napari, specific Python scripts…) For this purpose, the facility is equipped with several GPU workstations for efficient processing of big data, available to users.
