Master internship IRIMAS: Fibers as Optical Resolution Test-patterns for Unlabeled Nanoscopy Assessment

Position Information:
Context
IRIMAS develops Tomographic Diffractive Microscopy (TDM), a state-of-the-art method in
unlabeled optical microscopy, in terms of resolution (<150nm) and measure of sample
optical properties. Resolution is however difficult to measure, as no dedicated 3D test pattern is
available, sometimes leading to dubious claims. Objects like beads or 2D USAF test patterns are often used, but not good to evaluate resolution along the optical axis. Inspired from superresolution methods such as PALM/STORM, which can use microtubules to test their ultimate 3D resolution, we propose using electrospinned nanofibers in TDM. LPMT introduced the technique in France, and masters the process through home-made setups, but also now owns a commercial system. 
LPMT laboratory will prepare for IRIMAS appropriate test samples (diameter of the fibers, density of deposition), and conversely, our setup will be used to characterize optical properties (index of refraction, birefringence) of electrospinned nanofibers.
The internship
IRIMAS proposes a Master 2 internship to develop specific test patterns to characterize IRIMAS TDM system using nanofibers from LPMT:
- Preparation of nanofibers at LPMT, varying fiber diameter and composition, and the density
of fibers, from isolated ones to “thick” samples composed of several layers of fibers; - Use of a Nanolive system and of the IRIMAS TDM set-ups to image the samples;
 - Development of specific image processing methods to quantify resolution;
 - Improvement of the present image reconstruction methods: in particular, implementation of
advanced image reconstruction methods for thick, multi-diffractive samples; - Characterization of micro- and nanofibers optical properties.
 
The relative importance of these items can be adapted depending on the candidate’s cursus.
