Technician Lightsheet microscopy

Dr. Serge van de Pavert’s team uses light sheet microscopy to analyze immune cells in complete tissues, such as embryos or tumors. This state-of-the-art imaging technology acquires complete tissues in record time and allows a reliable and fast 3D reconstruction of the samples. In this context, the technician will participate in a research project aiming at developing, adapting and implementing clearing and immuno-fluorescent staining techniques on different tumor samples from mouse models. This project is carried out in close collaboration with teams from the CIML, the Institute of Developmental Biology of Marseille (IBDM) and the Institute of NeuroPhysiopathology (INP) who will provide tumor samples (melanoma, sarcoma, lymphoma, pancreatic tumor, glioblastoma). The technician will organize the sample collection, including dissection, and performs the staining and clearing protocols. The technician will perform the experiments under the supervision of the team’s permanent engineer. The technician will also assist in the genotyping of the different transgenic mouse lines of the laboratory and in the management/organization of the laboratory.
Activities
-Organ preparation, sectioning (cryostat, vibratome), staining and acquisition by confocal microscopy
-Sample preparation, staining and clearing for light sheet microscopy
-Computer analysis of results (Zen software, Image J/ACE, Imaris for 3D imaging); presentation of results.
-Genotyping of mouse colonies
-Animal experimentation
-Participation in the collective life of the laboratory (cleaning, orders, stock management).
Skills required
-General knowledge of immuno-fluorescence.
-Mouse breeding and genotyping; practice of animal experimentation, specifically in mice
-Confocal microscopy
-Proficiency in office automation (Office software) is essential
-Good understanding of written and oral scientific English

Education
-Bac + 2
-Animal use certification is a plus.
Salary is based on technician (assistant-ingénieur) with less than 3 years of relevant experience.
