Description of the training activity

Objectifs
1. Rappeler et/ou fournir les éléments de base de la microscopie par FLIM : qu’est-ce que c’est le FLIM ? Le FLIM est-il la bonne méthode pour répondre à ma question ?
2. Établir des « lignes-guide » pour tout utilisateur et/ou personnel de plate-forme souhaitant démarrer des expériences de microscopie par FLIM : quel type d’échantillon ? Pour quelle application ? Comment bien préparer son échantillon ? Quels sont les limites des échantillons ? Quels sont les contrôles indispensables à intégrer pour chaque expérience et quoi faire si les contrôles présentent des problèmes ?
3. Illustrer, avec des ateliers pratiques, des applications en FLIM pour la biologie qui sont susceptibles d’être l’objet d’une large diffusion au sein des laboratoires de recherche et des plateformes de microscopie : comment déterminer la présence d’interactions protéine-protéine à l’échelle sous-cellulaire (par ex. à la mitochondrie ou dans le cytoplasme des cellules), la concentration de certains métabolites intracellulaires (par ex. NAD(P)H, FAD) et l’activité d’une voie de signalisation intracellulaire (kinase ; autophagie)

➢ Axe 1 : principes du FLIM
➢ Axe 2 : préparation d’échantillons et contrôles des expériences en FLIM
➢ Axe 3 : FLIM pour déterminer les interactions protéine-protéine
➢ Axe 4 : FLIM en mode biosenseurs (activation de voies biochimiques intracellulaires)
➢ Axe 5 : FLIM en mode autofluorescence (activation de métabolites intracellulaires)
